I submitted an application to an interdisciplinary program and specifically mentioned only one faculty member whom I am especially interested in.  I have just gotten an email from another faculty member thanking me for expressing interest in his lab and wanting to Skype about my application, experience, interests, etc.  To be honest this professor's lab looks really cool and is more focused on a lot of stuff I did in undergrad research than any one else in the program, but I definitely never mentioned his lab in my SOP.  His email does not say anything about him being on the admissions committee either.  How do I handle this?

If you're interested in being in this lab, reply enthusiastically and say you'd be happy to Skype with the prof. Don't read too much into the email, it's probably just a form email he sent everyone who he is interviewing.
