Is it bad that I like answering my own questions? My PI thought it was funny that he doesn't even need to answer my questions anymore because I typically just answer them myself =.=? wtf

Hi All, I have a question regarding salting out technique using ammonium sulfate. The pI of my protein of interest is between 4.9-5.3. I noticed there were a lot of DNA and RNA in my protein sample after my initial purification via amylose affinity chromatography. I know that I could use DNase and RNase to get rid of them, but at this point I do not have the funding to get neither of them. I have to come up with alternative ways to approach this. Hmm, I was wondering if I could use 30% ammonium sulfate to crash out my protein and leave other DNA/RNA behind. Would this work? Would both DNA/RNA co-crash out with m protein of interest? My protein is currently in a buffer containing 10 mM of maltose and is at pH 7.6. My PI recommended using desalting column to separate them so I should also give that a try.

No, they evaluated your application as a Masters applicant and not a PhD applicant. However, you may ask them to see if they could re-evaluate your application as a PhD applicant, but I'm not sure about funding situation though. They might not have enough of funding for an extra spot. -

One of the PhD schools I got accepted to offer me a conditional acceptance letter. They wanted me to send them a copy of my Masters' transcript, a pdf copy of my thesis and other materials from the register's office to prove that I have completed my Masters successfully prior to start my doctoral program. One of my colleagues from my cohort class got accepted because she had a Masters degree as well but she never finished it. It made her look real bad when she tried to apply to join a research group. -
You could use Khan Academy to refresh your memory, they have some very useful clips on there.

You could mention it in your personal statement, and discuss about the stuff you worked on briefly. It would be difficult to tell if it would count or not though.

I had over 4.5 years (2 from undergrad and 2.5 from Masters) of research experience before I applied for PhD programs. I think 1.5+ years should be okay. You also want to stay in the same lab the whole time, unless it's rotation. Because it would make you look real bad when Admission Committee evaluating your application, it would make them think that you either: 1) hard to work with 2) lack of commitment 3) someone doesn't know what he wants 4) lack of maturity 5) inconsistent Your chance might be higher because you have a Masters degree depends on if it was thesis-based or course-based. You could 1) volunteer in a lab for a year to gain more hands-on research experience or 2) work as a technician in a lab or a company, while work your personal statement and GRE exam

There are some programs that will take you in as long as you meet the minimum criteria, however, most of them do not offer any financial support for students, you are most likely will have to pay everything out of your own pocket. -
I ain't mess around, 2 papers will be submitted by the end of this summer.

I think at this point you should really re-evaluate yourself. If you are having so much trouble in your Masters program right now, do you think you'll do well in a PhD program? Even if you pass all the classes and other requirements to graduate, do you think you'll be competitive enough to compete with other people with the same degree? -
Are you sure that by having a high GRE score alone would get you into a decent program? I think your score is fine, you need to focus on publications if you don't have any, or relevant research experiences. For most of biological/physical science programs, the admission committee tends to focus more on your LoRs, SoP, and other accomplishments because they know that most of the GRE questions are pretty irrelevant to the scientific community.
